The kinetics of polyphosphate decomposition in water was studied using beta-glycerophosphate (beta-GP) as an inducer of polyphosphate-accumulated bacteria. Polyphosphate was completely decomposed by beta-GP and polyphosphate-accumulated bacteria. The concentration of polyphosphate was increased with increasing beta-GP concentration. When the initial concentration of polyphosphate was 100 microM, complete degradation was obtained at the concentration of beta-GP of 40 microM. The reaction was rapid, reaching half-consumption of polyphosphate in 0.5 min at 0.1 mM beta-GP, whereas it took 50 min at 3 mM beta-GP. The amount of glycine phosphate released was proportional to the concentration of the inducer, at least to 10 microM, and the concentration of glycine phosphate corresponded to half of the concentration of beta-GP. The amount of polyphosphate decomposition by beta-GP and polyphosphate-accumulated bacteria increased with increasing initial concentration of polyphosphate. The amount of glycine phosphate released was almost proportional to the initial polyphosphate concentration. The amount of glycine phosphate accumulated in the cells also increased with increasing initial concentration of polyphosphate. The increase of glycine phosphate in the cells of the methanogenic bacteria was similar to the amount of glycine phosphate released from polyphosphate.
